When you hit OK, you'll go back to the Define Sites interface again. When you click Done, Dreamweaver should pop up the Site window for the site.
Go to
Site -Connect
This should show you the current contents of your www/ directory on the web server. From here, you have a number of options, depending on how current the files on your hard drive are. Since I'm assuming that the files on your local hard drive are the most current for the site, I recommend you go to:
Site -Synchronize...
Choose Entire (your current) site from the Synchronize: selector and select Put newer files to remote from the Direction selector. Make sure that the Delete remote files not on local drive option is UNCHECKED (very important), then click on OK.
After your local machine thinks for a little bit, it should present you with a list of files that it's found that need to be synchronized. Assuming that the list is correct, just click on OK. You can uncheck the box next to a file to keep from uploading it, should you so choose.
Your machine will then upload all the files that it needs to in order to synchronize the local and remote copies of your site. Once it is complete, click on the Close button in the Synchronization dialog box and go check the site with your web browser. Everything should work just fine at this point.
